Design: PCR primers of 20-26 bp were designed in exons encompassing intronic regions of about 400 bp for 48 low-copy nuclear loci, with an annealing temperature arounf 60 C. PCRs of all nuclear loci were carried simultaneously on 48.48 IFC plates following Fluidigm protocol. Amplicons were then pooled and barcoded by sample for paired-end (2x300 bp) Illumina MiSeq.

The dataset was produced to infer the evolutionary history of four diploid Aegilops spp. (Ae. tauschii, Ae. umbellulata, Ae. caudata and Ae. comosa) and of four of their derivative allopolyploid species (Ae. cylindrica, Ae. crassa, Ae. geniculata and Ae. triuncialis). The dataset includes 420 accessions of these eight species, 29 replicates and 26 outgroup species: all other extent diploid Aegilops spp., Triticum monococcum, Triticum urartu, Secale cereale, Hordeum vulgare and Taeniatherum caput-medusae.
